FERTILIZATION OF MOUSE OVA IN VITRO

Summary.: Certain factors which could influence the process of fertilization in explanted ampullar parts of the oviduct were studied by a modification of the method of Brinster & Biggers (1965). The effects of homologous serum, glycine, lactate, glucose, pH and oxygen were tested, and also the fertilizing capacity of spermatozoa from the uterus 1 to 7 hr after copulation. The effect on fertilization of varying the concentration of non-agglutinated spermatozoa was evaluated. A decrease of glucose concentration produced a negative effect on fertilization and the initial development of eggs. A lower pH (6·8 to 7) was followed by lower fertility only in those ova which were fertilized 16 to 17 hr after injection of HCG. Fertilizing capacity was also lower in spermatozoa isolated from the uterus 61/4 to 7 hr after copulation. Concentrations of spermatozoa from 300 to 4500/mm3 had no effect on the percentage of ova fertilized.
